Description:
Poze 30mg Tablets contain Pioglitazone hydrochloride, an oral antidiabetic medication used in the management of type 2 diabetes mellitus. It belongs to the class of drugs known as thiazolidinediones, which work by increasing the body’s sensitivity to insulin, thereby helping to control blood sugar levels. Pioglitazone does not increase insulin secretion but improves the body’s response to it, making it particularly useful in patients who are insulin resistant.

Indications:

• Type 2 diabetes mellitus, especially in patients who cannot be adequately controlled by diet and exercise alone
• May be used as monotherapy or in combination with other antidiabetic agents (e.g., metformin, sulfonylureas, or insulin)

Mechanism of Action:
Pioglitazone activates the peroxisome proliferator-activated receptor gamma (PPAR-γ), a nuclear receptor that regulates the transcription of insulin-responsive genes. This results in improved insulin sensitivity in adipose tissue, muscle, and the liver.

Dosage and Administration:

• The usual starting dose is 15mg or 30mg once daily, with or without food
• Dose may be adjusted based on clinical response, up to a maximum of 45mg daily
• Regular monitoring of blood glucose and HbA1c is recommended

Side Effects:

• Common: Weight gain, edema, headache, muscle pain
• Serious: Heart failure, liver dysfunction, risk of bladder cancer (rare with long-term use), bone fractures in women

Precautions and Warnings:

• Not recommended in patients with active liver disease or heart failure (NYHA Class III or IV)
• Use with caution in patients with history of bladder cancer or at risk of fractures
• Regular liver function tests are advised
• Not for use in type 1 diabetes or diabetic ketoacidosis
