This binding prevents the incorporation of isoleucine into bacterial proteins, disrupting their synthesis. Mupirocin also has the ability to inhibit DNA replication and RNA transcription, further contributing to its bactericidal activity. It achieves this by interfering with DNA gyrase and topoisomerase IV, essential enzymes involved in bacterial DNA replication and maintaining DNA structure. These dual actions make mupirocin effective against a range of gram-positive bacteria, including methicillin-resistant Staphylococcus aureus (MRSA) and other staphylococcal species.
